PriboFast® M200
DETERMINATION OF DOXYNIVALENOL IN CEREAL GRAINS BY HIGH-PERFORMANCE LIQUID CHROMATOGRAPHY (HPLC) AOAC Official Method PVM 2:1997
EXTRACTION
1. Weigh out 25 g of ground sample into a 250 mL glass blender jar or 250 mL Erlenmeyer flask.
2. Add 100 mL of 84+16 acetonitrile-water.
3. Blend at high speed for 2 minutes (blender) or shake for one hour on gyratory shaker.
4. Filter through fast qualitative filter paper and pipet about 10 mL of the filter extract into a 15 x 85 mm test tube.
PURIFICATION
5. For each sample use a new PriboFast® M200.
6. Remove plunger from column to be used.
7. Place column inside test tube for collection and place test tube in test tube rack.
8. Pipet appropriate sample extract volume into top of PriboFast® column. This volume will be determined by the method instructions.
9. Use plunger to quickly push sample through the column purification material into the test tube.
10. Remove column and dispose of in proper receptacle.
11. Reject into the LC or evaporate the purified extract to dryness per method instructions.
12. Reconstitute the dried sample per method instructions and perform the chromatography analysis.

• Column: Kromasil, 4.6×15cm, C-18
• Temperature: room temperature.
• Mobile phase: (5+5+90) Methanol+Acetonitrile+Water
• UV: 220nm
• Flow rate: 1 mL/min.
• Injection volume: 20ul
